上皮稳态失衡与肿瘤
1. 上皮极性/稳态调控与肿瘤
聚焦上皮极性基因的功能机制,系统解析以下四方面的细胞生物学问题:(1)极性信号在上皮稳态维持中的核心作用;(2)极性信号与亚细胞结构功能调控(例如线粒体和细胞骨架)的关系以及在上皮细胞能量代谢中的作用;(3)具有双定位功能的极性信号行使入核调控功能的重要意义;(4)探究基质刚度、组织压力等力学微环境破坏上皮细胞功能、并促进肿瘤进展的力学生物学机制。通过以上四方面的研究,解析极性蛋白丢失协同致癌突变驱动肿瘤发生的代谢重构以及肿瘤微环境调控的机制。
2. 硒稳态失衡与肿瘤发生
深入研究微量元素硒及硒蛋白家族因子在肿瘤氧化代谢压力下的多层次调控规律,通过线粒体蛋白调控细胞死亡、炎症以及与免疫代谢中全新机制探讨,试图解析肿瘤代谢重编程以及表观遗传调控规律,以明确其在肿瘤恶性转化中通过对肿瘤细胞衰老等方式实现改善肿瘤免疫微环境的免疫抑制作用,将为硒抗肿瘤免疫干预提供新的临床治疗干预方案和新靶点。
3. 上皮稳态与肝胰代谢性疾病
以肝胰上皮屏障为核心,研究上皮屏障紊乱与糖脂代谢微环境的相互调控机制,解析慢性代谢疾病中的免疫 - 代谢串扰规律,揭示肝屏障功能异常介导的以下病理生理过程:脂肪干细胞趋化分化等功能失衡、代谢紊乱与慢性免疫代谢微环境的关系及其炎症的分子基础,从而发掘慢性肝病及代谢综合征防治的新靶标与新策略。
